Binding of nerve growth factor receptor in sympathetic ganglia.

چکیده
(125)I-Labeled nerve growth factor binds to membrane preparations from superior cervical ganglia of rabbit in a specific fashion indicative of an interaction with the physiological receptor. Of various tissues examined, binding was observed only in the superior cervical ganglia, a presumed target organ of nerve growth factor. Binding of labeled factor is displaced by nonradioactive factor but not by several other peptide hormones. Binding to ganglia of preparations of nerve growth factor treated with N-bromosuccinimide closely parallels their biological activity. Binding of nerve growth factor receptor is saturable with an affinity constant of about 0.2 nM, similar to the reported plasma levels of the factor. Nerve growth factor does not elicit insulin-like biological effects in isolated adipocytes, and it does not modify the binding of [(125)I]insulin or [(125)I]epidermal growth factor to fat cells or liver membranes.

Binding characteristics and apparent molecular size of detergent solubilized nerve growth factor receptor of sympathetic ganglia.
Nerve growth factor (NGF), a hormone-like regulator of sympathetic neuron ontogeny and metabolism affects its target cells initially by associating with specific plasma membrane receptors. We have solubilized the NGF receptor of adult rabbit superior cervical ganglia (SCG) with the nonionic detergent Triton X-100. The high-affinity equilibrium binding constant of the detergent-extracted recepto...
متن کاملSpecific ablation of the transcription factor CREB in sympathetic neurons surprisingly protects against developmentally regulated apoptosis.
The cyclic-AMP response element-binding (CREB) protein family of transcription factors plays a crucial role in supporting the survival of neurons. However, a cell-autonomous role has not been addressed in vivo. To investigate the cell-specific role of CREB, we used as a model developing sympathetic neurons, whose survival in vitro is dependent on CREB activity. We generated mice lacking CREB in...
متن کاملSympathetic Gangliogenesis and Temporo-Spatial Glycoconjugates’s Terminal Sugars Distribution
Lectin binding histochemistry was performed on the developing sympathetic ganglionic cells to investigate the distribution and density of defined carbohydrate terminals on the cell surface glycoproteins during autonomic system morphogenesis. Sprague-Dauley rat embryos from 9th gestational day to birth were fixed and paraffinized. Serial sections of these specimens were incubated with different ...
متن کاملAbsence of the p75 neurotrophin receptor alters the pattern of sympathosensory sprouting in the trigeminal ganglia of mice overexpressing nerve growth factor.
Sympathetic axons invade the trigeminal ganglia of mice overexpressing nerve growth factor (NGF) (NGF/p75(+/+) mice) and surround sensory neurons having intense NGF immunolabeling; the growth of these axons appears to be directional and specific (). In this investigation, we provide new insight into the neurochemical features and receptor requirements of this sympathosensory sprouting. Using do...
متن کاملNerve growth factor receptors on chick embryo sympathetic ganglion cells: binding characteristics and development.
NGF is essential for the development and maintenance of sympathetic and certain sensory neurons. The NGF receptors on the surface of sympathetic ganglion cells from chick embryos were characterized; they consist of high-affinity receptors with a dissociation constant of about 10(-11) M, and low-affinity receptors with a dissociation constant of about 10(-9) M. There are more than 10 times as ma...
